Study on the stability of leucomalachite green (LMG) in the matrix of aquatic products

摘要: 孔雀石绿(malachite Green,MG)是影响水产品质量安全的重要风险隐患,其进入水生动物体内后,会代谢为隐色孔雀石绿(leucomalachite green,LMG),MG及其代谢物LMG在水产品中的稳定性又受到诸多因素影响。为探究基质品种和保存条件对LMG稳定性的影响,本研究以鲢(Hypophthalmichthys molitrix)和鲤(Cyprinus carpio)为研究对象,采用液相色谱-串联质谱法测定LMG回收率,考察LMG回收率在不同基质品种(鲢和鲤)中随贮存时间(1~5个月)的变化,探究加标基质的LMG回收率在不同贮存温度(22~23℃室温、2~6℃冷藏、-12℃和-20℃冷冻)下随贮存时间的变化,同时考察了基质称样和加标时间间隔对LMG回收率的影响。结果表明:1)基质品种是影响LMG测定结果的重要因素。在相同的冷冻贮存条件下,贮存1~5个月的鲤基质中,LMG的回收率基本稳定在90%~110%之间,而冷冻贮存3个月的鲢基质,LMG添加回收率仅为30.7%。2)空白基质贮存时间和基质加标后的贮存温度和时间是影响LMG测定结果的另外两个重要因素。新鲜的鲢和鲤基质对LMG测定结果影响较小,空白基质贮存时间越长和加标后基质样品的保存温度越高、保存时间越长,LMG的添加回收率越低。3)称样与加标间隔时间对LMG的回收率无显著影响。上述结果说明,在开展水产品中MG残留量检测的能力验证或实验室进行质量控制时,应尽可能选择新鲜的鲤基质样品,并尽快进行检测;如果样品不能及时检测,则需冷冻保存,并尽早安排检测。本研究可为实验室内部质量控制及以添加方式开展能力验证和数据比对提供参考。Abstract: Malachite green (MG) is an important risk affecting the quality and safety of aquatic products, and its stability is affected by many factors. Malachite green is metabolized into leucomalachite green (LMG) after entering aquatic animals. In this paper, in order to investigate the effects of breeds and preservation conditions on the stability of malachite green, these factors were studied in Hypopththalmichthys molitrix and Cyprinus carpio by the liquid chromatography-tandem mass spectrometry (LC/MS/MS) based on LMG recovery. The silver carp and carp samples were operated and preserved in different conditions by adding LMG solution, and the dependences of LMG content on time were studied. The results showed that the matrix breed was the most important factor affecting the results of LMG. Carp matrix frozen for 1-5 months had little effect on the recovery of LMG with the recovery of 90%-110%, while silver carp matrix frozen for 3 months had great influence with the recovery of 30.7%. The storage time of blank matrix and the preservation conditions of matrix after adding LMG standard were another two important factors affecting the recovery of LMG. Fresh silver carp and carp matrix had little effect on the results of LMG. However, the long storage time of blank matrix, and the high preservation temperature and long storage time of matrix after adding LMG standard caused a low recovery of LMG. The time interval from weighing sample to standard addition had not obvious influence on LMG recovery. When carrying out competency verification or quality control in laboratories, fresh carp matrix samples should be a better choice which will be tested as soon as possible. If the sample cannot be detected in time, it needs to be frozen and tested as soon as possible. This study provides references and technical supports for laboratory internal quality control and capacity verification by external adding method.
