Influence of Eluent Polarity on Separation of Sediment Long-Chain Alkenones in Silica Gel Column Chromatography

【目的】研究洗脱溶剂极性对海洋沉积物中C37和C38烯酮的柱层析分离效果及对重建古海表温度的影响,探寻能完全洗脱长链烯酮而不引入过多杂质的洗脱溶剂配比。【方法】通过改变正己烷(H)与二氯甲烷(D)体积比来调节洗脱液极性,按极性从小到大对标准样品进行洗脱,并计算每种烯酮分子的洗脱量。【结果】当VHVD=8∶1时烯酮开始被洗脱出,到1∶2时可被完全洗脱。烯酮C38:2 Et最先被洗脱出,而C37:3最后被洗脱出。各烯酮分子被洗脱量的峰值出现在不同极性的洗脱液,按极性排序为C38:2 Et < C38:3 Et < (C38:2 Me、C37:2< C37:3。【结论】洗脱液极性对不同烯酮分子洗脱效果有差异,如果C37洗脱不完全,可能会使偏大,导致换算的温度偏高。VHVD小于1∶2时,烯酮分子可被完全洗脱。

【Objective】This study aims to explore the influence of elution solvent polarity on the separation of C37 and C38 alkenones in marine sediments by column chromatography and its effect on reconstructed sea surface temperature, to find the solvent ratio that can completely elute all C37 and C38 alkenones without introducing too much impurities.【Method】 In this study, the polarity of the eluent is adjusted by changing the ratio of n-hexane (H) and dichloromethane (D).The standard samples are eluted with increasing polarity, and calculate the eluted amount of each alkenone.【Result】Alkenones start to be eluted when VHVD=8∶1, and can be completely eluted when VHVD=1∶2.C38:2 Et is the first to be eluted, and C37:3 is the last.The maximum percentage of each eluted alkenones appears in the eluent with different polarities, and the order of the eluent polarity is C38:2 Et < C38:3 Et < (C38:2Me, C37:2) < C37:3.【Conclusion】 The results reveal different elution effects of elution solvent polarity on alkenones.If the C37 is incompletely eluted, may be higher than the real value, resulting higher calculated temperature.When the ratio of H∶D is less than 1∶2, alkenones can be completely eluted.
