Protective effect of phillygenin (PHI) on liver injury induced by microcystins (MCs) in loach (Paramisgurnus dabryanus)

  • 为探究连翘脂素(phillygenin,PHI)对亚慢性毒性浓度微囊藻毒素(microcystins,MCs)诱发的大鳞副泥鳅(Paramisgurnus dabryanus)肝脏损伤的保护作用,对体质量为(100.13 ±5.23 )g的泥鳅腹腔注射剂量为17 μg/kg(体质量,下同)MCs 1 d后,分别连续注射3 d不同剂量的PHI(累计剂量0.3、0.6、1.2、2.4 mg/kg),第6天时取泥鳅肝脏样品,比较肝脏组织病理、谷丙转氨酶(ALT)活性、氧化损伤相关指标[丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)、谷胱甘肽还原酶(GR)]及炎性因子[白细胞介素1β(IL-1β)和肿瘤坏死因子α(TNF-α)]水平变化。结果表明:MCs可诱导泥鳅肝脏组织发生病理损伤;与对照组相比,MCs组泥鳅ALT活性、MDA含量及IL-1β和TNF-α两种炎性因子水平均显著升高(P<0.05),3种抗氧化酶活性显著降低(P<0.05);与MCs组相比,注射PHI能减轻MCs诱发的泥鳅肝脏组织病理损伤,显著降低肝脏中ALT活性、MDA含量及IL-1β和TNF-α两种炎性因子水平(P<0.05),显著提高肝脏中SOD、GSH-PX和GR活性(除0.3 mg/kg PHI剂量组外)(P<0.05)。研究表明,PHI能够减轻MCs引起的泥鳅肝脏组织病理与生理功能损伤、氧化损伤和炎症反应,对MCs所致肝脏损伤具有保护作用。
