MS-222对大黄鱼幼鱼麻醉效果、组织结构及抗氧化酶活性的影响

Effects of MS-222 on anaesthesia, tissue structure and antioxidant enzyme activity of juvenile large yellow croaker (Larimichthys crocea)

摘要

为探究MS-222长时间麻醉对大黄鱼(Larimichthys crocea)幼鱼机体的影响,采用不同质量浓度(10、20、30、40、50、60、70 mg/L)的MS-222,对体质量为(97.97±9.56)g的大黄鱼幼鱼进行24 h麻醉试验,通过对肝脏、鳃丝和脑的组织形态及超微结构变化进行显微和电镜观察,评估长时间麻醉对幼鱼机体的影响;通过对肝脏抗氧化酶活性和血清皮质醇浓度的检测,分析麻醉对机体抗逆性的影响。结果表明:在(16±1)℃条件下,用质量浓度为40 mg/L的MS-222麻醉时,幼鱼可进入深度镇静期,用60~70 mg/L的MS-222麻醉时,幼鱼可进入深度麻醉期,但用70 mg/L的MS-222麻醉24 h后,幼鱼的复苏率仅为50%;组织切片观察显示,用40 mg/L的MS-222麻醉24 h时,对机体可能存在轻微损伤;生理生化试验显示,用20 mg/L的MS-222麻醉时,幼鱼血清皮质醇浓度在1.5 h时显著上升(P<0.05),用20、40 mg/L的MS-222麻醉时,总体上能增强幼鱼肝脏中的超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)活力,在清水中复苏6 h后,SOD、CAT、GSH-Px活力基本能恢复至空白组水平。研究表明,40 mg/L的MS-222可作为本试验条件下大黄鱼幼鱼长时间运输的适宜质量浓度。

为探究MS-222长时间麻醉对大黄鱼(Larimichthys crocea)幼鱼机体的影响,采用不同质量浓度(10、20、30、40、50、60、70 mg/L)的MS-222,对体质量为(97.97±9.56)g的大黄鱼幼鱼进行24 h麻醉试验,通过对肝脏、鳃丝和脑的组织形态及超微结构变化进行显微和电镜观察,评估长时间麻醉对幼鱼机体的影响;通过对肝脏抗氧化酶活性和血清皮质醇浓度的检测,分析麻醉对机体抗逆性的影响。结果表明:在(16±1)℃条件下,用质量浓度为40 mg/L的MS-222麻醉时,幼鱼可进入深度镇静期,用60~70 mg/L的MS-222麻醉时,幼鱼可进入深度麻醉期,但用70 mg/L的MS-222麻醉24 h后,幼鱼的复苏率仅为50%;组织切片观察显示,用40 mg/L的MS-222麻醉24 h时,对机体可能存在轻微损伤;生理生化试验显示,用20 mg/L的MS-222麻醉时,幼鱼血清皮质醇浓度在1.5 h时显著上升(P<0.05),用20、40 mg/L的MS-222麻醉时,总体上能增强幼鱼肝脏中的超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)活力,在清水中复苏6 h后,SOD、CAT、GSH-Px活力基本能恢复至空白组水平。研究表明,40 mg/L的MS-222可作为本试验条件下大黄鱼幼鱼长时间运输的适宜质量浓度。
